Aqueous Buffers

The same buffer every time

Downstream processing of monoclonal antibodies and other recombinant proteins requires large volumes of buffer to maintain pH and conductivity during chromatographic purification steps and to stabilize proteins during hold steps and processing. YMC’s Buffer In-line Dilution (BID) systems with digitally controlled, diaphragm metering pumps and optimized fluidic design deliver a precise and accurate flow of both buffer concentrate and diluent. This ensures better control of final buffer composition and minimizes the variation of critical quality attributes, such as (ionic strength, concentration, pH, and conductivity).

Types of dilution systems

Standard, pre-engineered, or custom-designed systems

YMC can provide a standardized platform BID system, a customized system, or a BID functionality built into our standard chromatography system to help you:

  • Produce consistent point-of-use or centralized buffers
  • Automate complex dilution processes
  • Minimize operator exposure to potential chemical hazards
  • Reduce labor costs associated with buffer preparation
  • Reduce the footprint of your tank farm by using smaller concentrate bags or tanks
  • Reduce CIP volume and waste
  • Increase scheduling flexibility by enabling the use of buffer concentrates in convenient single-use bags

Please ask us to share specific examples of the reliability and efficiency gains we’ve helped our customers achieve with our BID systems.

An exclusive design delivers unrivaled performance

The BID system includes our proprietary digital control, pump technology, optimized system design, and sensors to monitor the dilution process. System software and automation allow the user to configure and run recipes that accurately and precisely blend the constituent solutions to produce ready-to-use buffers. Our BID system:

  • Automatically prepares accurate and precise buffers at point-of-use or a central location
  • Enables dilution factors of up to 50:1 with ≤ ±1% variation in flow accuracy and precision
  • Performs robust and simple buffer dilution based on control with volumetric flow rate to ensure consistent buffer ion composition
  • Offers a fluidic design that facilitates mixing, reduces the time required to reach set points, and enables drainability
  • Includes user-adaptable software that is recipe-driven with configurable, pre-programmed sequences that minimize operator intervention.

Features

BID for downstream processes using central systems or built into your chromatography skid:

  • Liquid-to-liquid precision blending
  • Point-of-use dilution from concentrates
  • In-line dilution as an integral feature of GMP EcoPrime LPLC systems
